This preprint establishes a standardized mixed lymphocyte reaction platform to evaluate MSC immunosuppressive potency, using MSCs cultured with 5% Human Platelet Lysate (HPL) from PL BioScience before testing.

This preprint establishes a standardized mixed lymphocyte reaction (MLR) platform for evaluating the immunosuppressive potency of mesenchymal stromal cells (MSCs), identifying HLA-DR modulation, preferential CD8+ T-cell suppression and PD-1/PD-L2 checkpoint signaling as functional readouts of a mechanistic potency axis.

Both adipose- and bone marrow-derived MSCs used in this platform were cultured with 5% Human Platelet Lysate (HPL) from PL BioScience GmbH, Germany, before potency testing. HPL formed part of the culture foundation behind this standardized potency assessment approach.
